Overview
Gene expression is the process by which the information encoded in a gene is converted into a functional product, principally through transcription of DNA into messenger RNA and translation of that mRNA into protein. Its regulation determines which genes are active, in which cells, and to what degree, governing development, differentiation, and cellular responses to physiological and pathological signals. Study of gene expression spans the analysis of expression levels in normal versus diseased tissue, as exemplified by altered VCAM-1 and apoptosis-related gene activity in colorectal and other cancers, and the dissection of mechanisms that drive resistance to therapy. Regulatory layers include transcription-factor binding, illustrated by analyses of regulatory single-nucleotide polymorphisms in genes such as STAT4 and EPAS1, and epigenetic control, as seen in the regulation of BDNF in chronic pain. Post-transcriptional regulation by microRNAs, which fine-tune target genes and serve as quantifiable diagnostic markers, is a recurrent theme, alongside the developmental roles of conserved gene families such as Hox genes. Research in this area quantifies and compares expression patterns, identifies the molecular controls behind them, and applies expression signatures to disease diagnosis, prognosis, and the understanding of cellular function across health and disease.
